Toxic PR Poly-Dipeptides Encoded by the
C9orf72 Repeat Expansion Target LC Domain Polymers
Prof. Ilmin Kwon of the Department of
Anatomy and Cell Biology Sungkyunkwan University School of Medicine and Steven
L. McKnight of the Department of Biochemistry UT Southwestern Medical Center
and their team used two complementary approaches in search of intracellular
targets of the toxic PR poly-dipeptide encoded by the repeat sequences expanded
in the C9orf72 form of amyotrophic lateral sclerosis (ALS).
ALS, also known as Lou Gehrig¡¯s disease that
cause the extinction of motor neuron of the central nervous system of the
patients is the reason of stiffen and shrunken muscle and finally patients die
because of stiffen respiration muscle. According to several recent studies,
about half of ALS is derived from C9orf72 mutation.
Evidence is presented indicating that low
complexity (LC) sequences represent the direct target of PR
£î
binding
and that interaction between the PR
£î
poly-dipeptide and LC domains is
polymer-dependent. These studies indicate that PR
£î
–mediated toxicity may result from
broad impediments to the dynamics of cell structure and information flow from
